Instruments

2.1 Introduction

The basic requirements for a confocal scanning optical microscope (CSOM) are point illumination, point detection, a confocal lens system, and a method of scanning the image. A number of different optical systems have been used to fulfill these requirements. They range in complexity from simple microscopes, using a laser, pinhole, and a scanning stage, to more complicated instruments employing white light sources, galvanometer mirrors, acousto-optic cells, or Nipkow disks for scanning. In this chapter we will discuss a few variations of the CSOM. The different approaches will be compared, pointing out the most common applications for each instrument.
