This chapter describes the mechanism-based screening for the discovery of novel antifungals. Antifungal agents were identified by the ability to inhibit fungal growth in agar solidified media. There are a number of variations on this basic assay design, including the Walksman streak assay, agar overlay method, and disk or agar plug diffusion assay.
