Attenuation in Depth

Attenuation of the ﬂuorescence emission intensity with increasing depth due to

absorption and scattering is a common problem encountered in ﬂuorescence

microscopy. This effect is most speciﬁcally relevant to three-dimensional (3-D)

ﬂuorescence imaging (see Chapter 14), where serial optical sections are acquired

along the axial plane to obtain volumetric data stacks. Correction of loss of

intensity with depth is essential for both visualization and quantitative analysis

of data. However, correction of intensity loss is an inverse problem that has no

unique solution ...
