Before iOS 5, you always had to plug in your USB cable to your Mac or PC to sync new media and to back up your iPhone. Now, you only need to plug it in one more time, and just long enough to enable Wi-Fi Sync. In the Summary tab of iTunes, under Options, turn on the Sync with This iPhone Over Wi-Fi checkbox. Once you enable it, you can disconnect your iPhone (let the current sync finish first) and, from that point on, whenever you connect your iPhone to a power source, it will begin syncing with your computer via Wi-Fi.
