Polymer therapeutic refers to nano-sized (5–100 nm) polymer-based pharmaceuticals. It includes rationally designed macromolecular drugs, polymer-drug and polymer-protein conjugates, polymeric micelles containing covalently bound drug, and polyplexes for DNA delivery.
Currently, the major uses of polymer therapeutics are drug targeting delivery and controlled release.
Drug toxicity can be reduced by increasing the drug solubility through conjugation of a hydrophilic polymer scaffold to the drug in question. With conjugation to a polymer, drugs can also be protected from degradation, resulting in improved efficacy due to increased drug circulation times.
